Chemistry of photoproteins as interface between bioactive molecules and protein function

چکیده
Abstract: Bioluminescence of Okinawan squid includes a photoprotein named "Symplectin" which gives light by addition of Na+ (or K+). The chromophore is covalently bound-imidazopyrazinone analog to protein. Symplectin was extracted with 0.6M KC1 and was analyzed to be composed of 3 different molecular-size proteins. Symplectin A, the smallest photoproteins, was purified by gel permeation chromatography to have 15kd by MALDVTOF mass spectrometry. The molecular mechanism of this luminescence is described. Bioluminescence is also applied for trace analyses and the chromophore in some new fireflies is chemically analyzed. We made a very sensitive detection system of luciferin to a detection limit of 10-'6 moY1Opl by using immobilized luciferase and a single photon-counter. An example of the firefly luminescence application is demonstrated in an inhibitory-assay of protein phosphatase activity by inhibitors in the same class of tautomycidokadaic acid.

Background: During pregnancy, the maternal immune system is in close contact with cells and tissue from the semiallogenic fetus .The Human Leucocyte Antigen (HLA) class Ib molecules, HLA-E, -F and -G, are expressed at the materno-fetal interface. Because of the apparent immunoregulatory functions of these proteins, they may be involved in successful acceptance of the semi-allogenic fetus during...

Appearing as a computational microscope, MD simulation can ‘zoom in’ to atomic resolution to assess detailed interactions of a membrane protein with its surrounding lipids, which play important roles in the stability and function of such proteins. This study has employed the molecular dynamics (MD) simulations, to determine the effect of added DMPC or DMTAP molecules on the structure of D...

X-ray diffraction (XRD) technique due to it's strength for providing visualized atomic model of molecules, stands as first in the field of characterization of new synthesized molecules. Here, the analysis of diffraction data on discrete units of new, not yet fairly known structure of molecule $rm C_{52} H_{60} N_4 S_8 Zn_2$ is performed to obtain a quantitative view of the structure of the syst...
متن کاملPrediction of In Silico ADME Properties of 1,2-O-Isopropylidene Aldohexose Derivatives
Retention behavior of molecules mostly depends on their chemical structure. Retention data of biologically active molecules could be an indirect relationship between their structure and biological or pharmacological activity, since the molecular structure affects their behavior in all pharmacokinetic stages. In the present paper, retention parameters (RM0) of biologically active 1,2-O-isopropyl...
